GADS constitutively interacts with SLP-76. This interaction is mediated by the C-terminal SH3 domain of GADS and a 20 amino acid proline rich region in SLP-76. Upon LAT phosphorylation, GADS interacts with LAT Y171 and Y191 via its SH2 domain. This interaction connects SLP-76 and LAT. The LAT-GADS-SLP-76 complex creates a platform for the recruitment of multiple signaling molecules, including PLCgamma1, GRB2, NCK, Rho GEFs, VAV and the Tec-family kinases ITK and BTK.
