Stabilisation of RAS:RAF by 14-3-3 14-3-3 has been displaced from Ser259. 14-3-3 may now bind its higher affinity Ser621 (S2) site. This stabilises an 'open' Raf-1 conformation that is catalytically active, and bound to p21ras via both p21ras-binding domains, CRD and RBD.
