Nephrin, is tyrosine phosphorylated by the Src family tyrosine kinase, Fyn in developing and injured podocytes. Phosphorylation of three of these tyrosines (1176, 1193 and 1217) results in the formation of a preferred binding motif (YDXV) for the SH2 domain of the adaptor protein NCK, which is one of the bricks linking Nephrin to the cytoskeleton. Phosphorylation of tyrosine 1158 results in a binding motif for the p85 regulatory subunit of PI3K.
