Cleavage of NOTCH1 by ADAM10/17 at the S2 cleavage site, which should precede the S3 cleavage by gamma-secretase, has not been studied in the context of NOTCH1 activation by CNTN1. NOTCH activation by CNTN1 is deltex-dependent, but the exact mechanism for action of the NOTCH:DTX complex has not yet been elucidated.
