TSC2 function is affected in at least two ways: first, phosphorylation decreases the activity of TSC2; second, phosphorylation destabilizes the TSC2 protein. This destabilization is achieved by disrupting complex formation between TSC1 and TSC2 and inducing ubiquination of the free TSC2 (PMID 12172553). Phosphorylation of complexed TSC2 by PKB induces degradation of both TSC1 and TSC2 through the proteosome pathway (PMID 121676664). Phosphorylation of complexed TSC2 by PKB may therefore result in the dissociation of the TSC1:TSC2 complex (PMID 12423332).
